Research
Abstract
The main objective of our research is the discovery of new reactions of synthetic and mechanistic interest. This exploratory program is complemented by target-specific syntheses. In this regard, selected natural products that are structurally amenable to function as bioreductive alkylating agents, and thus potential anticancer agents, are of particular interest. In addition, we are interested in the design and synthesis of non-natural products that will selectively cleave DNA but only subsequent to an activating chemical step.

Problems of current interest are the following: (1) synthesis and chemistry of un usual ketenes, e.g. 1; (2) generation and chemistry of unique diradicals such as 2 and bis-quinonemethides 3 as well as o-quinonemethides; (3) synthesis of DNA-cleaving agents such as the silylquinones 4 and 4-alkynyl-4-hydroxycyclobutenones 5; (4) synthesis of naturally occurring quin-ones having structural features as potential bioreductive alkylating agents, e.g., griseusin A; and (5)exploration of a new tandem oxy-Cope/transannular ring closure to polyquinanes 7 and the use of this reaction to synthesize natural products in this series.
