Abstract for “Discovery of BMS-986458, an Orally Bioavailable BCL6 Ligand-Directed Degrader” - Natalie Holmberg Douglas (BMS): 

Targeted protein degradation (TPD) is an emerging therapeutic approach that harnesses the cell's own disposal machinery to destroy disease-driving proteins, including many long considered "undruggable." One such target is BCL6, a transcription factor that drives tumor growth and treatment resistance in B-cell lymphomas like DLBCL and follicular lymphoma. Bristol Myers Squibb has disclosed two BCL6 degraders: BMS-986458, an orally available candidate now in phase I/II clinical trials, and BCL6-760, a tool compound for preclinical studies. This talk will cover the discovery of BMS-986458 and the challenge of advancing these large, "beyond rule of 5" degraders into the clinic.
